Iodine test is basically done to detect the pressence of starch in a particular part of a leaf in certain conditions. If you cover a part of leaf with a black paper and you leave another leaf as it is, after some days you will observe that the part which was covered does not show the pressence of iodinw where as the part of leaf which was not covered will show the pressence of starch. This is because the covered part used all its stored starch as it did not recive the sunlight where as this is vice versa in the case of the uncovered leaf, I hope it is clear!
