•To test the action of saliva on flour, take two test tubes with water and flour in it. Mix them well.

•After mixing add saliva in one test tube and keep it for incubation for about 30 minutes. Now add drops of iodine solution in both the test tubes.

•You will observe that blue colour is seen in test tube where saliva was not added. While blue colour won't be retained in the test tube with saliva.

•This observation was seen because saliva contains amylase enzyme. Amylase enzyme can break glycosidic bonds formed between carbohydrates.

•Since flour contains carbohydrate namely starch, it will be broken into it's monomeric components and hence blue colour of iodine is not seen.
