Materials Required :
Leaf, Compound microscope , Brush, China dish with water, Blade, Saffrain solution, Glycerine, Slide, Cover slip.
Procedure :
1. Take out the lower epidermis of the leaf with the help of blade.
2. Put it in China dish containing water.
3. Now put some saffrain drops in the China dish contains the lower epidermis.
4. Leave it for some time (about 15-20 minutes).
5. Take it out on slide with the help of brush from the china dish.
6. Apply Glycerine on the slide (over the leaf part).
7. Gently put cover slip on it by avoiding bubbles.
8. Take it under microscope and adjust it properly to see the stomata.
Observation :
We observes that the lower epidermis of leaf contains tiny opening called stomata. Where which each tiny opening have two guard cells.
