Peel off a leaf from half a piece of onion and using the forceps, pull out a piece of transparent onion peel (epidermis) from the leaf. Put the epidermis in the watch glass containing distilled water. Take a few drops of safranin solution in a dropper and transfer this into another watch glass.

To mount the onion peel on the slide.
Material required:
Safranin, cotton, onion, cloth and glycerin.
Instrument and laboratory
Light Microscope
Slide, forcepe, glass, coverslip, knife, needle.
Procedure:
1] Cut the onion and remove it's peel from the inner surface of onion.
2] Put the peel in a watch glass containing water.
3] Add few drops of safranin stain into the water glass to stain the peel.
4] Remove the peel with the help of camel hair brush and put it on clean slide alongwith few drops of water and provide the shape by cutting it.
5] Wash extra stain with water and remove excess water with the help of filter paper.
6] Put the drop of glycerine on the slide over the peel as required and clean the excess with filter paper.
7] Now put coverslip over the peel with help of needle.
8] The temporary mount of onion peel is ready for observation in under the microscope.
precautions:
1] Slide is clean and dry .
2] Peel should be mounted in the center of slide.
3] Use always brush to transfer peel from watch glass to slide.
4] Avoid the entry of air bubbles in inside during mounting.
Note:
Wash hands during experiment and under the supervision.
