It should be kept in the sunlight . the level of water in the beaker should be over the level of stem of the funnel. while removing test tube to prevent escape of gas evolved and remove gently . Observation : you will notice that in the place of water gas is filled .

Ensure that the level of water in the beaker is above the level of the stem of the inverted funnel. Place the apparatus in the sun for at least 3 hours. After sometime it is observed that gas bubbles come from the hydrilla plants and these are collected at the end of the test tube.
