Aim - To observe nucleus in onion peel cell

Materials required - Onion peel , stain ( saffrain , methylene blue or red ink ) , slide , cover slip , filter paper and microscope

Procedure -

  • Take a small fleshy portion of onion bulb and break it into pieces to obtain the thin translucent membrane
  • Now keep the membrane on the slide and add a drop of saffrain or methylene blue for staining
  • Cover the cover slip and leave it for few minutes
  • Add few drops of water from one side of cover slip such that extra stain is washed away
  • Use filter paper for soaking extra water
  • Observe under microscope

Observation - A dark spot can be observed towards the periphery which is nucleus . Thick boundry of cell called as cell wall is also visible

Inference - Onion peel consists of dense round body called nucleus

Precautions - Tap the cover slip to remove air bubbles
