On basis of reaction of bacteria to the Gram's stain. In this process bacteria are stained with Crystal violet then washed with alcohol ,then safranin the universal stain is used on bacteria and the gram negative bacteria don't react to stain but the stain reacts with gram positive bacteria thus this shows the difference.
