Hapten is usually defined as a substance that is not immunogenic as such but can bind to antibodies of the appropriate specificity (hapten–specific antibodies). A hapten can be made immunogenic by coupling it to an immunogenic protein (the carrier). The conjugate is then immunogenic not only with respect to carrier epitopes but also to the hapten. ‘Antigenic’ is sometimes incorrectly used as a synonym for ‘immunogenic’, but free hapten is ‘antigenic’ and not ‘immunogenic’. Antibodies specific to a hapten can only be raised when the hapten is foreign to the individual to be immunized.
