- A section of the leaf is covered on both sides black paper, preventing sunlight from reaching the leaf region beneath the black paper.
- The plant is now exposed to direct sunshine, but only a portion of the leaf is visible.
- After a few hours, the covering is removed from the specific leaf. The black sheets are removed, and the leaf is then decolorized in boiling water with alcohol. Decolorization happens when the chlorophyll in the leaf is eliminated, causing the leaf to lose its pigmentation.
- Iodine solution is applied to the decolorized leaf.
