Procedure:

  1. For 1-2 days, place a healthy green potted plant in a dark room. This is done to ensure that the plant consumes all of its stored food and that the leaves are starch-free.
  2. A section of the leaf is covered on both sides black paper, preventing sunlight from reaching the leaf region beneath the black paper.
  3. The plant is now exposed to direct sunshine, but only a portion of the leaf is visible.
  4. After a few hours, the covering is removed from the specific leaf. The black sheets are removed, and the leaf is then decolorized in boiling water with alcohol. Decolorization happens when the chlorophyll in the leaf is eliminated, causing the leaf to lose its pigmentation.
  5. Iodine solution is applied to the decolorized leaf.

Observation:

The part of the leaf that was covered by paper remained colourless after adding the iodine solution, whereas the region that was exposed to sunlight turned blue-black.

Inference:
