This line " men may come and men may go but I go on forever"tells us that the life of a man is mortal and for a period of time but the life of a brook is immortal and eternal and it flows continuously .

This is the refrain in the poem 'men may come, men may go' but 'I go on forever'. The refrain in the poem suggesting what the brook things about its journey across various land scape. It also indicates the bravity of human beings is compared the longevity of brook.
In living beings also, a person can born grows into the child and become an adult. Then it reaches the stage of an old man and then dies.
The brook originates from its dwelling point where the small water birds and down the valley to meet the over flowing river.
The continuity ant external existence of brook is compared with the transitory nature of human. Despite, hurdles but the brook flows continuously. Similarly, the human beings should move on their way irrespective various ups and down, joys and sorrows, pains and grief and take these in their stride.
