- Tissue culture, also called as, callus culture, is a technique that utilizes the fragments of plants and cultures them in nutrient media under suitable artificial environmental conditions.
- Protoplast culture is the isolation of protoplasts by mechanical and enzymatic methods.

- The development of a plant in tissue culture depends on two fundamental principles of cells which include totipotency and plasticity.
- The application of protoplast culture includes the development of hybrids, cell cloning, developing genetically transformed plants and membrane studies.

- In tissue culture, segments of different plant parts, known as explants, are cultured. The explants that are used in tissue culture include leaf, stem, root, petiole, hypocotyl, cotyledon, embryo, or meristem.
