The differences between protoplast culture and tissue culture are as follows:

(i)

  • Tissue culture, also called as, callus culture, is a technique that utilizes the fragments of plants and cultures them in nutrient media under suitable artificial environmental conditions.
  • Protoplast culture is the isolation of protoplasts by mechanical and enzymatic methods.

(ii)

  • The development of a plant in tissue culture depends on two fundamental principles of cells which include totipotency and plasticity.
  • The application of protoplast culture includes the development of hybrids, cell cloning, developing genetically transformed plants and membrane studies.

(iii)

  • In tissue culture, segments of different plant parts, known as explants, are cultured. The explants that are used in tissue culture include leaf, stem, root, petiole, hypocotyl, cotyledon, embryo, or meristem.
  • The methods used to perform the protoplast culture include hanging-drop cultures, micro-culture chambers and soft agar matrix.
