The main difference between the rough and the smooth endoplasmic reticulum (ER), is that the rough endoplasmic reticulum have ribosomes in their membranes (and this is why they are also called granular while the smooth agranular- since they lack ribosomes on there membranes).
Another notable difference is tha the rough ER are mostly found in protein synthesizing cells while the smooth are found in lipid forming cells.
Function in membrane biogenesis:
Cell membranes are mainly made up off lipids and proteins. The ER manufacture the bulk of the lipids and membrane proteins responsible for making the cell membranes.
