The most basic difference between RER and SER is the presence of ribosomes. When ribosomes attach to the surface of an ER, it gives a characteristic rough appearance; hence it is called Rough ER. On the other hand, a smooth ER does not have ribosomes on its surface. It possesses ribosomes attached to its membrane.

SER has no ribosomal particles on the surface, hence look smooth. It helps in manufacturing of lipids and fat molecules. ... Thus, they help in membrane biogenesis.

difference btw. rough ER and smooth ER-

Rough ER has ribosomes attached to its membrane that is why is is called 'rough' whereas Smooth ER has no ribosomes attached to it..

Another difference is that Rough ER is formed from cisternae and a few tubules however SER is formed from vesicles and tubules

hope it helped.. good day!
