The main difference lies between the SER and RER is the presence of ribosomes, as SER do not have ribosomes but RER have ribosomes. SER mainly produce and export lipids and protein and function for detoxification, while RER produces, secrete and export proteins and few hormones.

RER=Rough Endoplasmic Reticulum.
It is having rinosomes attached on it that is why it is also known as Ribosomal endoplasmic reticulum.
Presence of ribosomes makes its surface rough which also helps in the proteins syynthesis.
SER=Smooth Endoplasmic Reticulum .
smooth endoplasmic reticulum donot have ribosomes attached on it so they donnot help in protein synthesis but they help in lipids synthesis which help in building cell membrane.
