The leaf peel is so lightly colored that it becomes difficult to see under a microscope. It's transparent. Staining it makes it darker and highlights it properly so that stomata can be seen easily. Now take the example of a glass. Sometimes you dash into a glass door by mistake since it is transparent. Had the glass door been colored you would have easily seen it and would not have made the same mistake. Staining makes it easily visible. Hope you understand
