“ELISA” is used to ‘detect viruses’, where “Alkaline phosphatase is the key reagent”.  

The most common substrate for ELISA is that alkaline phosphatase which is “p-Nitrophenyl Phosphate (pNPP)” that gives a ‘yellow product’ that is measured at the wavelength of 405 nm.

The principle behind ELISA (“Enzyme Linked Immunosorbent assay”) is that the unknown protein is immobilised and linked to an antibody, which in turn is linked to an enzyme. This when incubated along with the substrate gives the quantification.
