Submerge it in a solution of baking soda which will serve as a source of carbon dioxide  

Invert the glass funnel and cover the aquatic plant  

Fill the test tube with water and cover the opening of the test tube with your thumb so there is no air confined inside while submerging the test tube.  

Then invert the test tube and then gently submerge it and cover the stem of the glass funnel as seen in the diagram

Then keep the setup in a sunlit area or under a bright light  

Carefully remove the test tube without entering any air  

Test the gas by a glowing splinter and observe.
