The presence of starch in leaves can be tested by Iodine test. When we remove chlorophyll from leaf by boiling it in alcohol and then put two drops of iodine solution, its colour change to blue indicates the presence of starch.

  • The required item to conduct this test are Beaker, alcohol, Iodine solution, tests tube, and a leaf.
  • The principle theory is whenever Iodine reacts with starch, then blue-black colour is obtained.
  • Take a green leaf in the test tube and add an alcohol solution in the test tube.
  • Now heat the beaker which is having water using a  burner, place the test tube in the beaker.
  • So the leaf should be boiled in water.
  • After that, the boiled leaf placed in 90% of ethanol.
  • With time the leaf will start losing it's green color and will become colorless.
  • Now take out the leaf from the test tube and add the iodine solution on the leaf.
  • You will find the colour of the leaf changes into blue-black.
  • Hence, it is proved that the leaf contains starch in it.
