There was no Prime Minister of India before Indo-Pak division but Jawahar Lal Nehru was the head of Interim government formed on September 2, 1946. The Nehru was head of interim government under the title of Vice-President of Viceroys Executive Council but played a similar role of a Prime Minister until Indo-Pak division or Independence.
After Independence Nehru became the first Prime Minister of India.
