Totipotency, that is, the full range of developmental capabilities, requires that all the genetic information be in place in the cell nucleus. In the past, nuclear transplantation has been an effective tool to assess the general mode of cellular differentiation (i.e., whether cells become specialized through the physical loss of genetic material). Currently, nuclear transplantation shows that epigenetic changes occurring in cells undergoing differentiation can be reversed, at least in a minority of the cells used as nucleus donors.
