TISSUE CULTURE
Tissue culture is growing plant cells in an artificial medium under sterile conditions is called as tissue culture.

let's see meaning of some terms which we use in tissue culture
explant: part of a plant taken and grown in testtube by application of nutrient medium.
Totipotency: the capacity of an explant to generate whole plant from it is called as totipotency.

Tissue culture technique was first initiated by Gottlieb Haberlandt in 1902. He is considered as father of tissue culture.
BASIC TECHNIQUE OF PLANT TISSUE CULTURE.
1)PREPARATION OF SUITABLE NUTRIENT MEDIUM. Sutable nutrient medium as per objective of culture is prepared and transferred in to suitable containers.
2)SELECTION OF EXPLANTS. Selection of explants such as shoot tip should be done. this is preffered because, plants cultured through explants obtained from meristem will give disease free plants(Reason-mersitems divide more rapidly than virus do.)
3)STERILISATION OF EXPLANTS. Surface sterilisation of explants by disinfectants (e.g., sodium hypochlorite) and then washing the explants with sterile distilled water is essential
4)INOCULATION. transfer of explants into the suitable nutrient Medium (which is sterilised by filter-sterilised to avoid microbial contamination) in culture vessels under sterile conditions is done.
5)INCUBATIONS. Growing the culture in the growth chamber or plant tissue culture room, having the appropriate physical condition ( i.e., artificial light atleast for 16 hours), temperature( -26° C) and relative humidity (50-60%) is required.
6) REGENERATION. Regeneration of plants from cultured plants tissues is carried out.
7)HARDENING. Hardening is gradual exposure of plantlets to an evironmental condition.
8)PLANTLET TRANSFER. After hardening, plantletsplantlets are transferredcto the green house or field conditions.
