  • Agrobacterium tumefaciens is called genetic engineer.

  • Because some part of its Ti plasmid called T-DNA is transferred and incorporated into host cell DNA where it is expressed along with the host DNA.

  • Any gene contained in T-DNA therefore becomes part of host genetis system.

  • If the plasmid in the bacterium does not have an insert, the presence of a chromogenic substrate gives blue coloured colonies.

  • The presence of insert results into insertional inactivation of the beta-galactosidase and the colonies do not produce any colour.

  • Such colonies are marked as recombinant colonies.
