a) DNA cannot pass through the cell membrane as it is not lipid soluble and since cell membrane is bilipid layer it won't allow it's passage.

b) Proteases are used to break down the proteins to increase the ease of extraction of DNA.

c) Cloning vectors are useful for generating many copies of your gene. Expression vectors are associated with the actual expression of the gene into mRNA and protein in the target organism. Cloning vectors usually contain features associated with the insertion or removal of DNA fragments.
