There is a difference because CaCl2 absorbs the water vapour that is transpired from the plant. The lower surface transpires more than the upper surface. So when you weight the vials the vial in the lower part will be heavier than the vial in the upper part in the potometer.

(i) The CaCl2 vials are kept inside the cup to absorb water. (ii) After few hours, the weight of the CaCl2 vials will increase because they will absorb the water which is transpired by the leaf of the plant.
