to examine the starch accumulation in your  experimentally treated leaves ,you first need to remove green chlorphyll from them by using

green leaf

test tube

alcohol

container

water

procedure:
place leaf in a test tube and pour some alcohol in it.  Heat some water in a container and put test tube containing alcohol and green leaf into the container.chlorophyll dissolves in warm alcohol .Thus,leaf looses it green colour and become decolourise

To decolourise a leaf we have to remove green coloured chlorophyll from a leaf by boiling it first in water and then in alcohol
