How would you test the presence of starch in leaves?

The presence of starch in leaves will be indicated by a blue-back colour.

procedure:

  1. take a leaf of a potted plant
  2. Boil the leaf in methylated spirit or ethanol over a water bath till it becomes pale white due to removal of chlorophyll.

3.spread the leaf in a dish and add a few drops of tincture iodine / betadine solution on it.

observation:

the presence of starch will be indicated by a blue-back colour.

Take two potted plants of the same kind. Keep one in the dark for 72 hours and the other in sunlight. Perform the iodine test with the leaves of both the plants as given below. Now leave the pot which was earlier kept in the dark, undisturbed for 3 – 4 days and perform the iodine test again on its leaves.
