Number of nuclei is THREE in a fully developed male gametophyte.

Explanation:

In flower, the pollen grain is considered as male gametophyte and embryo sac is considered as female gametophyte. The formation of male gametophyte occurs through two stages namely Pre-pollination stage and Post-pollination stage.

As the pollen germination initiates in the pollen sac, it leads to the formation of large vacuole which causes the movement of nucleus to one side of the cell. The nucleus undergoes mitosis and this leads to formation of two daughter nuclei namely the smaller generative cell and the larger vegetative cell. In the post pollination stages the pollen tube develops and leads to complete male gametophyte with three nuclei namely one Tube nucleus and two male nucleus.
