We use saffranin and not glycerin because saffranin is used as a colouring substance and it is more convenient to observe as compared to a non coloured stomata under a microscope. Glycerin on the other hand can't provide any colour to the slide to be viewed.saffranin gives pinkish-red colour to a substance
