The last two lines are of poet's contemplation. Going through the title and tone of poem it seems that the poet regrets his decision of choosing the less traveled by road. This decision has made a big difference in his life.

In the last two lines of the poem, the poet accepts the reality. The poet made a choice and accepted the challenging path. He took the unexplored path in his life. He wanted to do something different in life so he choose the less travelled road. No, he doesn't regret his choice.
