Medical student here. It's been a couple years since pathology, but I'll try and recall. This was literally the first topic in the class, about cellular adaptation to stress, so forgive me that the details are scant.

Basically, intracellular stress (like stretching) will upregulate certain proteins that expedite cellular keratinization and proliferation. This leads to increased thickness of skin, because of higher numbers of dead cells, and increased thickness of each cell layer.

The calluses occur due to the rubbing of skin area, it may cause infection. It is formed due to divisions of keratinocytes and their accumulation on the outer skin. It can be removed by the keratolytic agent. Scrubbing of calluses at the time of foot spa is done to remove the dead skin cells that are creating pressure on live cells on skin that is causing pain.
