Steps to preparing temporary mount of human cheek cells.

Step one: Obtain cheek cells by gentle scrapping of the inside part of the mouth with a sterile cotton swab. after obtaining the cheek cells, place a drop of physiological saline to a clean microscope slide.

Step two: Smear the cotton swab on the center of the clean slide in order to get the cells at the center of the slide then add a drop of methylene blue to the smear and gently place a cover slip on the top.

step three: Place the slide on the microscope for observation to find the cells.

Step four: Use x4 or x10 magnification to view the cells.

1. obtain cheek cells by gentle rubbing of cotton swab on inner part of mouth.


2. now try to get cells on the centre of slide.


3. then add a drop of methylene blue


4. now  place a cover slip on the top.


5. observe it under microscope.
