In addition to the chromosomal DNA the mitochondria contains nucleic material which is called as the mitochondrial DNA. This ‘mitochondrial DNA’ is conserved for generations without destruction. It is this mitochondrial DNA which is used for phylogenetic mapping.  

In addition, the ‘mitochondrial DNA’ is inherited only from the mother, the sperm does not contribute mitochondria for the zygote development only the ovum contains the mitochondria.
