- Take 1 ml. of 1% Starch Solution in experimental tube (A) and control tube (B).
- Add 1 ml. of diluted saliva in test tube A and 1 ml. of water in control tube (B).
- Leave both the test tubes undisturbed for half an hour in a beaker maintained at 37°C.
- Now add a few drops of dilute iodine solution to both test tubes and note the colour change.
Iodine gives blue colouration with solution in test tube B but no blue colouration with solution in test tube A.
Salivary amylase (Ptyalin) enzyme of saliva has digested starch to double sugar maltose in tube A which does not give blue colour with iodine.
