Replication of DNA takes place during the S-phase or Synthetic phase of Interphase.

When a eukaryotic cell undergoes mitosis and cytokinesis for complete cell division, interphase follows for recovering the cell’s condition.

The DNA of that cell replicates during the S-phase, after rebuilding of the protein in the cell during the G1 phase.

The chromosomes attain a relaxed condition, where an enzyme called DNA polymerase helps to read the original DNA strands, for creating new DNA double helix with the same genetic traits.
