First method:

Negative sentences include “never, not and other words like this”. And positive sentence strictly don't use such words. So here are some positive forms of the given sentence :

That sentence was very hard for me to understand.

I found it difficult to understand your joke.

I am unable to understand your joke.

Second Method:

"I don't think I understand that joke".

If you want to have the positive form of the sentence which conveys the same meaning, the following sentence could adequately serve the purpose:

“That joke is rather difficult for me to understand.”

Third Method:

“What’s the joke?”

“I fail to understand that joke.”

“I am unable to understand that joke.”

“I think I fail to understand that joke.”

Go with the first one, others though grammatically correct are ludicrous.
