In order to check the respiration rate of the plants, it is necessary to use KOH filled test tube inside the conical flask.

As soon as plants start respiration process, the CO2 levels will increase, giving way for more volume of air to be infused. But as KOH consumes the CO2, the water levels will rise up as a vacuum has already been created.
