Hey there,
The enzymatic isolation of protoplasts can be carried out by two approches:
Two steps or sequential method: The tissue is first treated with pectinaze (macerozyme) to separate cells by degrading midlle lamella. One step or simultaneous method: This is the preferred method of protoplast isolation.
