First take the peel . Then place it on a transparent object like glass.add 2-3drops of water and enough stain.cover it with a cover slip.remove the air bubbles in the slide.adjust the microscope.and observe the slide through the microscope under 10x ,....etc

In preparing temporal slides of plant epidermis like onion, rheo, talinium slides of plant cell is prepared by adding various chemicals, cuts and pigments.

EXPLANATION:

At first few water droplets are added in slide to moisten it. Then take the onion or rheo cut and peel off it by using knife shaped tool. Then it is placed sharply on the slide.

Some drops of iodine solution should be added. Then glass cover slide is placed carefully. Then it is placed under the microscope. Then turn on the microscope light. Then look through the eye piece carefully and make observation.
