Iodine test is used to test for the presence of starch. When a plant is kept in the dark for 72 hours it is denied light which is a condition necessary for photosynthesis.The plant in the dark room will bend towards the hole and may come out of the hole. Plant needs sunlight for photosynthesis. If it gets no light it will be no longer able to function. plants are kept in dark for 72 hours before photosynthesis experiments because so plants will not able to perform photosynthesis and make food . In that condition plant will use energy present in leaves in form of starch.
