The distance increases from C to D along time, at the point D the particle is at a rest position. While from D to E, the particle covers negative distance means that the particle started moving to opposite direction. Hence the velocity of a particle is negative at the point E.

Instantaneous velocity of a particle is the slope of the tangent to the displacement - time graph at a point. At point E, the slope is negative.
Therefore , the Instantaneous velocity of the particle is negative at point E.
