To study stomata by making temporary slide of leaf peel

Here are the steps to prepare the slide

1. Tear the lower epidermis of the leaf

2. Add it carefully to a petridish full of water to wash it

3. Remove the peel with a brush and place it on a clean petri dish

4. Stain the peel using safranin

5. After staining place the peel on glass slide with the help of brush. See to it that it does not fold.

6. Add a drop of glycerine over the peel

7. Cover the peel with a cover slide and observe under a microscope. Avoid bubbles while covering.
