' And that has made all the difference.' means that he is getting to discover a new path which not many people have discovered.

In the last two lines of the poem, the poet accepts the reality. The poet made a choice and accepted the challenging path. He took the unexplored path in his life. He wanted to do something different in life so he choose the less travelled road. No, he doesn't regret his choice.
