Now, we measure mass in kg and energy in joul.Then E= mc^2 gives relation between joul and kg. c^2 is conversion factor .You may say that it is like relation between work(in joul) and heat (in calory) with joule’s constant as conversion factor. This is simplest way to look at the mass energy relation.

If you heat a body the thermal energy contained by the body increases . This increase in energy gives increase in mass of the body which can be calculated by Einstein’s mass energy relation.
