Ribosomes are present only over the surface of rough endoplasmic reticulum. SER is mainly made of vesicles and tubules while RER is made of cisternae and tubules. SER involved in the synthesis of glycogen, lipids, and steroids while RER engaged in the synthesis of proteins and enzymes.

Answer:
The main difference lies between the SER and RER is the presence of ribosomes, as SER do not have ribosomes but RER have ribosomes. SER mainly produce and export lipids and protein and function for detoxification, while RER produces, secrete and export proteins and few hormones.
