In synoptic knob, there are vesicles and those vesicles have a chemical named as Acetylcholine, it's a neuro-transmitter, and it's released at the Synapse.
Synaptic cleft is the region which is adjacent to next neuron, it's where the one neuron releases neurotransmitter molecules (Acetylcholine).

Although, the synapses gap or cleft is very important and creates nerve impulses.
