1. Lock up the mirror, and the optical viewfinder image goes away. You won't be able to see if your subject moves before you press the shutter button.

2. Lock up the mirror, and phase-detect autofocus (which relies on light it collects from the viewfinder path) can no longer track the subject, either.

3. On some cameras that have Live View, locking up the mirror may be part of Live View, but there could conceivably be a time limit (to avoid overheating the sensor).

4. On cameras that implement a short mirror lockup as part of the self-timer feature, once you trip the timer, you may have no further control over the precise moment at which the camera starts to take the picture.

5. Locking up the mirror may drain the battery. On my camera, which has a mirror lockup feature for sensor cleaning, you are not allowed to lock up the mirror unless the battery is fully charged (more or less). Presumably it would be a Bad Thing if the battery ran flat and the mirror came crashing down while you had some sort of cleaning tool stuck under the mirror and in position to strike the sensor.
