Gram Positive Bacteria

  • Peptidoglycan 70 -80 %
  • Single layered
  • thickening 20-80 nm
  • lipid less
  • which retain dye

Gram Negative Bacteria

  • Peptidoglycan 10-20%
  • Double layer
  • Thickening 8 - 12 nm
  • Lipid 20 - 30 %
  • do not retain dye

Gram positive bacteria are those bacteria which can retain the crystal violet dye because of the thick layer of peptidoglycan and are stained purple with low resistance against antibodies and they can produce primary exotoxins and they have more resistance to drying and physical disruption. While  Gram negative bacteria on the other hand are stained red or pink as they do not retain the crystal violet dye because of thin layer peptidoglycan with more resistance against antibodies because of impenetrable membrane and they can produce primary endotoxins and they have low resistance to drying and physical disruption.
