Put the leaf in methylated spirits to remove the chlorophyll. Do the iodine tests for starch on the whole leaf. The part of the leaf not covered by the band paper turns a blue-black colour. The part of the leaf covered by the band does not turn blue-black because of lack of sunlight for photosynthesis to make starch.
