In order for replication to begin, the replication machinery must have access to the nucleotides that are encoded in the DNA strands. This access is granted through the enzyme known as DNA helicase.

The Function of DNA Helicase

Imagine for a minute that you are shopping and interested in a new pair of jeans. You want to try the jeans on, so you need to figure out how to get into them. The first thing you notice is the button. Opening the button gives a little room, but not enough for you to get into the jeans. You then pull down the zipper, which opens the jeans up even more, giving you the space you need to get into them.

This is very similar to the role that DNA helicase plays in DNA replication: DNA helicase is the zipper. It unzips our DNA strands to allow space for attachment and to expose the nucleotides that are used as a template during DNA replication and transcription.
