Materials required: Curd or yogurt (or any other source of lactobacillus), glass slide, cover slip, tooth pick, methylene blue stain, tissue paper, light microscope.
Procedure:
- Put a drop of curd on a clean test tube with the help of tooth pick.
- Smear it with the help of another slide.
- Put a drop of methylene blue on the smear and leave it for few seconds.
- Put the cover slip on the slide where smear is uniformly spread.
- One can remove excess stain with the help of tissue or paper towel.
- Observe the slide under light microscope; first under low power then in high power if needed.
Observation: They will appear as purple or blue colored rod shaped organisms.

ANSWER :
1) Take one or two drops of buttermilk on a slide , spread it on the slide .
2) Heat it slightly on a lamp for 3-4 seconds.
3) Add a few drops of crystal violet on it .
4) After 30 to 60 seconds , wash the slide gently with water .
5) Observe the slide under the compound microscope .
6) We will find a stick like structures , known as bacteria .
